Preliminary results of a new trial of a combination of rosuvastatin and Crestor have provided relief for patients who had recently lost their appetite. This trial was done following a double-blind, randomised trial to compare the effects of aripiprazole, simvastatin and Crestor, in patients with hyperlipidemia and aripiprazole, simvastatin or aripiprazole alone.
Crestor, a cholesterol-lowering medication, is the only statin that has been studied in humans. This study was conducted at a total cholesterol (TLC) of 0.02, 0.1 and 0.5 mg/ml, respectively. Patients were randomly allocated to receive either rosuvastatin, simvastatin or aripiprazole (25 mg daily) or dummy controls. The results of this trial show that, in patients with hyperlipidemia, the combination of rosuvastatin and Crestor (Crestor 10 mg) resulted in a clinically significant increase in total cholesterol and in low-density lipoprotein cholesterol (LDL-C) levels, whereas aripiprazole did not.
Aripiprazole is an alternative statin, available in tablet form as Abiraterone acetate or as Abiraterone-dihydrate. It was initially approved by the Food and Drug Administration (FDA) in 1999 and has been available in several generic forms since then. It is a relatively safe drug, with good tolerability and well-tolerability, with moderate-to-low-dose use. However, it is important to note that it is not effective in patients with hyperlipidemia and it can cause serious side effects, including lipid abnormalities and gastrointestinal disturbances.
Simvastatin is a calcium channel blocker and a statin, and it is a generic drug, as its active ingredient is simvastatin. Simvastatin is a brand-name drug that is manufactured by Pfizer. In the trials, aripiprazole produced a significantly lower LDL-C concentration than rosuvastatin or simvastatin. This result was confirmed in two additional studies.
Simvastatin was found to be as effective as rosuvastatin in reducing LDL-C concentrations and in preventing atherosclerosis, but it also produced a slightly greater increase in the rate of progression of atherosclerosis compared to rosuvastatin alone. In another study of patients with hypercholesterolemia, aripiprazole was found to be superior to rosuvastatin in reducing LDL-C levels and improving lipid profiles. These two studies were designed to compare the effectiveness of aripiprazole with aripiprazole alone and to demonstrate that aripiprazole produced a greater increase in LDL-C compared to rosuvastatin.
In conclusion, both aripiprazole and simvastatin produce statistically significant reductions in cholesterol levels when given in combination with either statin, as shown in the two studies.
In a second study, we studied the effect of aripiprazole on cardiovascular and lipid levels in patients with hypercholesterolemia. In this double-blind, randomised trial, the effect of aripiprazole was also studied in patients with hypercholesterolemia who had not responded to statin therapy. We found that patients with hypercholesterolemia who had responded to aripiprazole had higher cholesterol concentrations than those who had not.
This study is an observational study, and a further study is expected in the future. Patients with hypercholesterolemia or with a history of cardiovascular disease and/or lipid abnormalities were eligible to participate in the study. The study was conducted at the Hospital of General Medical Sciences, Lahore, Pakistan.
The study design is also observational, as patients were randomly allocated to take rosuvastatin (0.5 mg/kg) or dummy placebo, and to take rosuvastatin plus aripiprazole (25 mg) or dummy controls. There were two small-scale clinical studies in which the effect of rosuvastatin was compared to that of aripiprazole in patients with hypercholesterolemia and in patients with a history of hyperlipidemia.
